John Smith is a constituent - has his own record and comes to one particular event. Jane Smith comes to an agency tour but doesn't have her own record so is recorded simply as a participant. Major gifts person then has a conversation with Jane and wants to record the action.
Is it possible to link Jane's participant record to John's constituent record (where Jane is already listed as the spouse) and then enter the action on John's record but referencing Jane in the notes of the action?

I would make Jane a constituent in her own right. This conversation could be a prelude to a major gift so this person should be tracked as her own constituent not as an relationship to someobody else.
While it is possible to record Jane's action on John's record, it violates basic principles of database design. You should make Jane a constituent and record the action on her record.
